Effects of triptolide from Radix Tripterygium wilfordii (Leigongteng) on cartilage cytokines and transcription factor NF-κB: a study on induced arthritis in rats

چکیده
BACKGROUND Triptolide, an active compound of Radix Tripterygium wilfordii, is immunosuppressive, cartilage protective and anti-inflammatory both in human and animal studies of various inflammatory and autoimmune diseases, including rheumatoid arthritis, but its therapeutic mechanism remains unclear. The aim of this study is to investigate the effects of triptolide on cartilage cytokines in the CIA model. METHODS Sprague Dawley rats were immunized with type II collagen and orally administered with triptolide. The arthritic scores and incidence changes of the rats were observed. The expression of TNF-alpha, IL-6, COX-2 and NF-kappaB in paw cartilage was studied with immunohistochemical staining. RESULTS Triptolide, at both high and low doses, significantly lowered the arthritic scores, delayed the onset of arthritis and lowered the arthritis incidence. Triptolide treatment at both high and low doses lowered the expression of TNF-alpha, IL-6, COX-2 and NF-kappaB in paw cartilage in arthritic rats. CONCLUSION Triptolide lowers the arthritic scores, delays the onset of collagen induced arthritis and reduces the expressions of TNF-alpha, IL-6, NF-kappaB and COX-2 in paw cartilage in arthritic rats.

Triptolide protects bone against destruction by targeting RANKL-mediated ERK/AKT signalling pathway in the collagen-induced rheumatoid arthritis
Rheumatoid Arthritis (RA) is a chronic autoimmune disease characterized by the inflammatory synovitis and the destructive cartilage and bone. Triptolide is a biologically active component purified from the Chinese herbal plant Tripterygium wilfordii Hook F (TWHF), which has presented benefits for the treatment of RA. In the current study, we investigated therapeutic effects of Triptolide in col...

BACKGROUND/AIMS Triptolide (TPL), a main active ingredient of Tripterygium wilfordii has been shown to exert anti-inflammatory effect. The role of TPL on glomerular diseases remains unclear. METHODS This study aims to investigate the potential anti-inflammatory effect of TPL in rats with membranous glomerulonephritis (MGN). RESULTS Our data showed that the pathological kidney damage was sig...

BACKGROUND Tripterygium wilfordii Radix, a well-known traditional medicine in china which is used for treatment of inflammation, pain, tumor and immune regulation for centuries in china, accompany with the serious toxic side effects. This study was carried out for simultaneously analyzing the four main components (triptolide, triptophenolide, demethylzeylasteral and celastrol) in Tripterygium w...
متن کاملTriptolide inhibits osteoclast formation, bone resorption, RANKL-mediated NF-қB activation and titanium particle-induced osteolysis in a mouse model.
The RANKL-induced NF-κB signaling pathway is required for osteoclast formation and function. By screening for compounds that inhibit RANKL-induced NF-κB activation using a luciferase reporter gene assay in RAW264.7 cells, we identified triptolide (PG490), as a candidate compound targeting osteoclast differentiation and osteoclast-mediated osteolysis. Triptolide (PG490) is an active compound of ...
متن کاملNeuroprotection by triptolide against cerebral ischemia/reperfusion injury through the inhibition of NF-κB/PUMA signal in rats
Triptolide, an active compound extracted from the Chinese herb thunder god vine (Tripterygium wilfordii Hook F.), has potent antitumor activity. Recently, triptolide was found to have protective effects against acute cerebral ischemia/reperfusion (I/R) injury through inhibition of cell apoptosis. However, the regulatory mechanism of the effect remains unclear. We hypothesize that the regulatory...
